Inhibition Of Bacterial Peptidoglycan, Muramyl Dipeptide-Induced Apoptosis By Indomethacin In Vitro And In Vivo.

Purpose. To demonstrate anti-apoptogenic activity of the cyclooxygenase inhibitor indomethacin (IndoM) against muramyl dipeptide (MDP)-induced apoptosis in rabbit cells and rabbit uveal tissue.
Methods. The anti-apoptogenic activity of IndoM against MDP was assessed in vitro by inhibition of cell death, suppression of intrinsic and extrinsic caspases and apoptotic DNA ladders. In vivo, inhibition of anterior uveitis, aqueous humor protein, caspase activity in iris/ciliary body tissue from MDP and MDP plus IndoM injected eyes.
Results. IndoM (100 µg/ml) inhibited cell death, caspases and apoptotic DNA in rabbit renal cells. Similarly, IndoM inhibited the signs and symptoms of uveitis, breakdown of the blood-aqueous barrier and induction of caspases in iris/ciliary body tissue.
Conclusion.  The results support the cytopathogenic effects of MDPs in rabbit cells and uveal tissue, demonstrate the anti-apoptogenic activity of IndoM, and support the idea that the beneficial effects of non-steroidal drugs against MDP-induced uveitis may be due in part to inhibition of apoptosis in uveal tissue.
